Immunocaptured M. avium subsp. paratuberculosis was transferred to bead-beating tubes and fixed in a bead ruptor (bead beater), which was operated at speed 6 for 40 s. The tubes were cooled to room temperature, and 10 μl of proteinase K (200 μg) was added and incubated first at 65°C for 20 min and then at 95°C for 10 min.
