A 15 ml overnight culture of Clostridial cells was collected by centrifugation, washed 3X with cold PBS, resuspended in 1 ml of cold PBS and lysed by three 30-second sonications using an ultrasonic homogenizer (sonicator) with the samples being kept on ice for at least two minutes between each sonication.
