Tissue segments of first-daughter cladodes of plants grown in growth chambers were harvested at the beginning of the night period. Spines of the cladode segments were removed and tissue was sliced in 1 to 2 mm sections and immediately homogenized in a medium containing 25 mm HEPES buffer, using a rotor stator (polytron) homogenizer (4–5 repetitions of 5 s each).
