The construct was transfected into Spodoptera frugiperda (sf-9) cells, single plaques were isolated, and high-titer stocks were generated. Insect cells were resuspended and thawed in buffer A and supplemented with a protease inhibitor mixture.  The cells were lysed with a rotor stator (polytron) homogenizer.
